This MAb reacts with the HLA-DRB3 antigen, a member of MHC class II molecules. It does not cross react with HLA-DP and HLA-DQ. HLA-DR is a heterodimeric cell surface glycoprotein comprised of a 36 kDa alpha (heavy) chain and a 28 kDa beta (light) chain. It is expressed on B-cells, activated T-cells, monocytes/macrophages, dendritic cells and other non-professional APCs. In conjunction with the CD3/TCR complex and CD4 molecules, HLA-DR is critical for efficient peptide presentation to CD4 T cells. HLA-DR antigens also occur on a variety of epithelial cells and their corresponding neoplastic counterparts. Monoclonal antibodies (MAbs) to liver cell processes are useful in the identification of hepatic carcinomas and normal organ structures. MAb HSA98 binds to human hepatocytes and the majority of human hepatocellular carcinomas (HCC's). In frozen sections, it stains hepatic cells and may be used as a marker of the liver. Cell preparations of hepatocellular carcinoma biopsies or cell lines are found to bind HSA98 on the cell surface. This MAb stains liver hepatocytes in frozen human liver sections and is positive on the cell surface of human liver carcinomas.Primary antibodies are available purified, or with a selection of fluorescent CF Dyes and other labels. This MAb reacts with the rod domain of human cytokeratin-19 (CK19), a polypeptide of 40 kDa. Its epitope maps between amino acid 312-335. CK19 is expressed in sweat gland, mammary gland ductal and secretory cells, bile ducts, gastrointestinal tract, bladder urothelium, oral epithelia, esophagus, and ectocervical epithelium. Anti-CK19 reacts with a wide variety of epithelial malignancies including adenocarcinomas of the colon, stomach, pancreas, biliary tract, liver, and breast. Perhaps the most useful application is the identification of thyroid carcinoma of the papillary type, although 50%-60% of follicular carcinomas are also labeled. CD45R, also designated CD45 and PTPRC, has been identified as a transmembrane glycoprotein, broadly expressed among hematopoietic cells. Multiple isoforms of CD45R are distributed throughout the immune system according to cell type. These isoforms arise because of alternative splicing of exons 4, 5, and 6. The corresponding protein domains are characterized by the binding of monoclonal antibodies specific for CD45RA (exon 4), CD45RB (exon 5), CD45RC (exon 6) and CD45RO (exons 4 to 6 spliced out). The variation in these isoforms is localized to the extracellular domain of CD45R, while the intracellular domain is conserved. CD45R functions as a phosphor-tyrosine phosphatase. This MAb reacts with all isoforms of CD45R expressed by all hematopoietic cells, except erythrocytes, having a higher level of expression on lymphocytes than on granulocytes (Workshop IV). Cytokeratin 8 (CK8) belongs to the type II (or B or basic) subfamily of high molecular weight cytokeratins and exists in combination with cytokeratin 18 (CK18). This MAb recognizes all simple epithelia including glandular epithelium, for example thyroid, female breast, gastrointestinal tract, respiratory tract, and urogenital tract including transitional epithelium. All adenocarcinomas and most squamous carcinomas are positive but keratinizing squamous carcinomas are usually negative. Recognizes 183 kDa protein with DNA-binding characteristics, which is identified as a myeloid specific antigen. It reacts with myeloid precursor cells and granulocytes in bone marrow. Its antigen appears to be restricted to M2 and M3 acute myelogenous leukemia (AML) subtypes. Markers of myeloid cells are useful in the identification of different levels of cellular differentiation. This MAb reacts with early precursor and mature forms of human myeloid cells. It is useful in the identification of myelogenous leukemias, distinguishing granulocytic sarcomas from lymphoid malignancies and also in the study of differentiation and transformation of human myeloid cells.
